Urinary Rat Alpha GST and Renal Injury

1 alphaGST1rats Alpha Glutathione S-Transferase is found in the proximal tubules of rat kidneys and constitutes 1-3% of the soluble protein. Alpha GST has been shown to leak from proximal tubule cells into the urine in response to injury. Urinary alpha GST quantification is a sensitive and specific assay for proximal injury assessment. Together with RPA-1 (collecting duct) and GST Yb1 (distal tubules) it provides a broad picture of nephron status from a single urine sample. Matching assays for in-vitro and human samples enable easy compaisons between experimental and clinical studies .

The test procedure is based on the sequential addition of sample, anti alpha GST - enzyme conjugate, and substrate to microassay wells coated with anti rat alpha GST. The resultant color intensity is proportional to the amount of alpha GST present in the sample. The assay range is 1.56-100 ug/L, equivalent to 9.75-625 ug/L in stabilized urine samples diluted 1:5 and 0-5000 ug/L in serum samples diluted 1:50. The total assay time is 2.25 hours.


Recent News on Rat Alpha GST

HESIRat Alpha GST was included in the ILSI HESI Nephrotoxicity Working Group project and its performance assessed compared to the traditional markers used in toxicity / safety testing. The results of this study have been presented at many conferences, most notably the Society of Toxicology meetings in 2007 and 2008.

In May 2008 the rat alpha GST data was submitted to the FDA / EMEA along with another 2 biomarkers under the VxDs qualification scheme. It is expected to be fully qualified early in 2009.
