Rat Collecting Duct – RPA-1 / PapX5C10 

The Renal Papillary Antigen 1 (RPA-1) antibody (clone PapX5C10) is specific to the collecting duct epithelial cells of the rat nephron. RPA-1 is a highly glycosated high molecular weight protein whose exact structure remains unknown. Argutus Medical have a number of collaborations active on this project.

The image below shows the staining with anti-RPA-1 in a healthy rat kidney. The secondary antibody in this case was AlexaFluor 595 goat anti mouse IgG (h&l). This image clearly demonstrates the specificity of anti RPA-1 for the rat collecting duct tubules.

The image below shows double staining with:

  • Anti-RPA-1 with AlexaFluor 595 goat anti mouse IgG1 as the secondary antibody.
  • PMII25E1: murine anti-human connecting tubule specific IgG1 antibodywith AlexaFluor 488 goat anti mouse IgG1 as the seconadry antibody.

The anti human antibody (PMII25E1) stains the corresponding tubule part in the rat kidney.

The Anti RPA-1 antibody is utilised in the Argutus Medical RPA-1 EIA enabling early signs of collecting dust injury to be detetected from urine samples.

The micrograph below shows a healthy rat kidney cross section stained with anti-RPA-1 (Red) and anti-Tamm-Horsfall Protein (Green). One can see the collecting ducts clearly staining red in this image.

The image below shows a close up of a section of the above micrograph. This further demonstrates that anti RPA-1 does not share any affinity for Tamm-Horsfall Protein.
